    Medium development strategies and scale down models for a high density high productivity cell line

    Medium Development at Regeneron continues to enhance fed batch culture productivity. These efforts have been enabled through the development of high throughput scale down models in shake flasks and the ambr® 250. Design of Experiment (DOE) approaches have been applied to optimize the operating conditions in the small scale models leading to performance for growth and titer that match benchtop bioreactor with no off-set. The development of these representative scale down models and our approach to medium development will be described. A medium development case study will be presented from a recent Regeneron fed batch process with a cell line achieving high cell densities and depleting the culture of key amino acids. The traditional medium development approach of supplementing the culture with the depleted nutrients was unsuccessful: high amino acid consumption rates required large amounts of amino acids resulting in significantly increased culture osmolality and reduced productivity. Leveraging high throughput culture systems and multifactor DOEs, multiple medium composition factors in combination were rapidly evaluated. Mathematical models relating medium input factors to process outputs are generated that allow for process optimization. Using this approach, a new feeding strategy was developed that limits increases in osmolality and yields titers approaching 10g/L in both the scale down systems and a process that has been implemented for clinical scale manufacturing of a monoclonal antibody

    Sector Agroalimentario e Integración Regional: una simbiosis necesaria para la Seguridad Alimentaria y Nutricional

    Through a documentary-descriptive and quantitative-analytical research, supported by calculation and interpretation of trade position and intraregional trade dynamics indicators, this work examines relationship between agriculture, regional integration and food security. For this reason, it analyzes the role played by agricultural variable in the context of integration agreements, characterizes elements that confer sensitivity to agri-food sector and conditions its special treatment within regionalism experiences, and evaluates current state of regional agri-food integration between 2001 and 2019 by comparing three Latin American schemes: ALADI, CAN and MERCOSUR. The central discussion argues that deepening of agri-food trade through economic integration offers opportunities to guarantee food security, as an instrument capable of supplying food in a stable and fluid way (facing turbulences in international markets) and with lower transaction costs. It is concluded that in Latin America there is no integration from the agri-food trade point of view and offers public policies recommendations for agricultural development, trade and strengthening integration.Mediante una investigación documental-descriptiva y cuantitativa-analítica, apoyada en el cálculo e interpretación de indicadores de posición comercial y dinámica del comercio intrarregional, el trabajo examina la relación existente entre agricultura, integración regional y seguridad alimentaria. Para ello analiza el papel desempeñado por la variable agrícola en el contexto de los acuerdos de integración, caracteriza los elementos que confieren sensibilidad al sector agroalimentario y condicionan su tratamiento especial dentro de las experiencias de regionalismo y evalúa el estado de la integración agroalimentaria regional entre 2001 y 2019 comparando tres esquemas latinoamericanos: ALADI, CAN y MERCOSUR. La discusión central argumenta que la profundización del comercio agroalimentario a través de la integración económica ofrece oportunidades para garantizar la seguridad alimentaria, como instrumento capaz de suministrar alimentos de manera estable y fluida, frente a las turbulencias en los mercados internacionales y con costos transaccionales más bajos. Se concluye que en América Latina no existe integración desde el punto de vista del intercambio agroalimentario y se ofrece una serie de recomendaciones a nivel de políticas públicas en aras del desarrollo agrícola, del comercio y del fortalecimiento de la integración

    Siguiendo una investigación documental-descriptiva y cuantitativa-analítica, el artículo analiza, desde una visión retrospectiva, los principales rasgos que definen las tres primeras décadas del MERCOSUR y sus transformaciones. Enfatiza el estudio del desempeño comercial del proceso de integración durante el período 2000-2021, tanto en sus relaciones intrarregionales como extrarregionales, para aportar una visión prospectiva al análisis de su presente y futuro económico inmediato. También describe las principales tendencias del intercambio de bienes, los temas de reciente discusión y los últimos acontecimientos del relacionamiento externo MERCOSUR-Asia. Entre otras cosas, demuestra la pérdida de importancia comercial de la Unión Europea y Estados Unidos; su desplazamiento por Asia y China como primer mercado del bloque sudamericano; y el ascenso del Sudeste de Asia como mercado emergente, donde Vietnam, Singapur, Malasia, Indonesia y Tailandia, principales economías de la ASEAN, han ganado importancia dentro del comercio global de bienes del MERCOSUR. Y con base en estas evidencias concluye que el presente y el futuro económico del MERCOSUR (en comercio, inversión y cooperación) se encuentran en Asia-Pacífico, razón válida para instar a la (re)definición de una Agenda Asia.Following a documentary-descriptive and quantitative-analytical investigation, the article analyzes, from a retrospective perspective, the main features that define the first three decades of MERCOSUR and its transformations. It also describes the main trends in the exchange of goods, the topics of recent discussion and the latest events in the MERCOSUR-Asia external relations. Among other things, it shows the loss of commercial importance of the European Union and the United States; its displacement through Asia and China as the first market of the South American bloc; and the rise of Southeast Asia as an emerging market, where Vietnam, Singapore, Malaysia, Indonesia and Thailand, the main ASEAN economies, have gained importance within the global trade of MERCOSUR goods. And based on this evidence, it concludes that the present and the economic future of MERCOSUR (in trade, investment and cooperation) is in Asia-Pacific, a valid reason to urge the (re) definition of an Asia Agenda

    Measurements of eye lens doses in interventional cardiology using OSL and electronic dosemeters

    The purpose of this paper is to test the appropriateness of OSL and electronic dosemeters to estimate eye lens doses at interventional cardiology environment. Using TLD as reference detectors, personal dose equivalent was measured in phantoms and during clinical procedures. For phantom measurements, OSL dose values resulted in an average difference of 215% vs. TLD. Tests carried out with other electronic dosemeters revealed differences up to +/- 20% versus TLD. With dosemeters positioned outside the goggles and when TLD doses were > 20 mu Sv, the average difference OSL vs. TLD was 29%. Eye lens doses of almost 700 mu Sv per procedure were measured in two cases out of a sample of 33 measurements in individual clinical procedures, thus showing the risk of high exposure to the lenses of the eye when protection rules are not followed. The differences found between OSL and TLD are acceptable for the purpose and range of doses measured in the survey.Postprint (published version
